Monoclonal antibody therapy: "model" experiments with toxin-conjugated antibodies in mice and rats.

چکیده
The advent of cell fusion techniques for producing monoclonal antibodies has stimulated world-wide effort in the search for antibodies with specificity for neoplastic cells. In parallel, a number of laboratories have attempted to devise ways of attaching cytotoxic agents to antibodies with the aim of generating potent anticancer agents from monoclonal antibodies that exhibit sufficient specificity for Cancer cells. The most successful ploy for arming antibody molecules has been to couple them to highly poisonous toxins such as abrin, from the jequirity bean, and ricin, from the castor bean. Abrin and ricin are glycoproteins comprising two polypeptide subunits, A and B, joined by a disulphide bond. The B-chain binds to galactose-containing molecules which are to be found on most cell surfaces and the A-chain is believed to penetrate the plasma membrane or the membrane of an endocytic vesicle and kill the cell by damaging ribosomes (reviewed in [ W

Introduction: Immunoassay procedures for detecting and determining opioids in blood and other biologic fluids are based on Monoclonal antibodies. In the present study, monoclonal antibody against Morphine was taken into account. Methods: Hybridoma protocol was used in order to produce the monoclonal antibody against morphine in mice. For this purpose, five 6–8-week old female BALB/c mice were...

Objective(s): Leishmaniasis is endemic in 88 countries. Amastigote forms of Leishmania are experts at exploiting host cell processes to establish infection. Monoclonal antibodies are key reagents used in the diagnosis of infectious and non-infectious diseases. The aim of this study was to produce monoclonal antibodies against axenic amastigotes of the Leishmania infantum strain in Iran.Material...
متن کاملMonoclonal and Polyclonal Antibodies Specific to Human Fibromodulin
Background: The unique expression of fibromodulin (FMOD) in patients with chronic lymphocytic leukemia (CLL) has been previously reported. Detecting FMOD in CLL patients using specific anti-FMOD mAbs might provide a promising method in detection, monitoring, and prognosis of CLL. Objectives: In this study, we aimed for producing specific antibodies agains...
متن کاملAntibody Response to Human Extracellular HER2 Subdomain Proteins in Mice
Background: In addition to passive immunotherapy using anti-HER2 monoclonal antibodies, active immunotherapy via HER2 targeting is an interesting approach to inducing specific anti-tumor immune responses. We have recently reported the immunogenicity of HER2 subdomains following DNA immunization and HER2 protein boosting. In the present study, we evaluated the immunogenicity of different HER2 ex...
